以下細(xì)胞處理方法及細(xì)胞說(shuō)明書(shū)僅供參考,具體操作還需要根據(jù)到貨時(shí)細(xì)胞密度,細(xì)胞生長(zhǎng)狀態(tài)等具體情況,酌情處理,如需要更詳細(xì)技術(shù)指導(dǎo),請(qǐng)及時(shí)電話(huà)或郵件聯(lián)系。

一.貼壁細(xì)胞
1. 客戶(hù)接收到細(xì)胞,用75%的酒精消毒(建議配制75%酒精的水是滅菌過(guò)的)培養(yǎng)瓶外部。
2. 肉眼觀察細(xì)胞培養(yǎng)基顏色,顯微鏡觀察細(xì)胞生長(zhǎng)情況,并對(duì)細(xì)胞進(jìn)行不同倍數(shù)拍照(建議收到時(shí)的培養(yǎng)瓶拍一張照片,顯微鏡拍收到時(shí)的細(xì)胞100X 200X 各一張)。
3. 顯微鏡觀察細(xì)胞,當(dāng)細(xì)胞融匯至80%左右(可以傳代的情況下),細(xì)胞應(yīng)該在37度培養(yǎng)箱預(yù)溫1-2h后再做處理。如未達(dá)到細(xì)胞傳代密度,培養(yǎng)瓶應(yīng)預(yù)留一部分培養(yǎng)基繼續(xù)培養(yǎng)。
4. 嚴(yán)格無(wú)菌操作,打開(kāi)細(xì)胞培養(yǎng)瓶。
5. 將細(xì)胞培養(yǎng)瓶?jī)?nèi)的培養(yǎng)基用吸管完全吸出(嚴(yán)禁直接傾倒),放入另一無(wú)菌容器中(建議換新的培養(yǎng)基培養(yǎng)細(xì)胞)。
6. 用PBS 2-3ml/次輕輕洗細(xì)胞表面,加入適量的0.05%胰酶-EDTA消化細(xì)胞顯微鏡下觀察細(xì)胞變圓,輕輕拍打培養(yǎng)瓶直到細(xì)胞脫落,加入終止液終止。
7. 1000rpm5min離心,重懸細(xì)胞。換新的細(xì)胞培養(yǎng)瓶,37℃5%CO2 培養(yǎng)。
二.懸浮細(xì)胞
1. 接收到細(xì)胞,用75%的酒精消毒(建議配制75%酒精的水是滅菌過(guò)的)培養(yǎng)瓶外部。
2. 肉眼觀察細(xì)胞培養(yǎng)基顏色,顯微鏡觀察細(xì)胞生長(zhǎng)情況,并對(duì)細(xì)胞進(jìn)行不同倍數(shù)拍照(建議收到時(shí)的培養(yǎng)瓶拍一張照片,顯微鏡拍收到時(shí)的細(xì)胞100X 200X 各一張)。
3. 嚴(yán)格無(wú)菌操作,打開(kāi)細(xì)胞培養(yǎng)瓶。
4. 將細(xì)胞培養(yǎng)瓶?jī)?nèi)的培養(yǎng)基用吸管完全吸出(嚴(yán)禁直接傾倒)。
5. 1000rpm5min離心,重懸細(xì)胞。 換新的細(xì)胞培養(yǎng)瓶和新鮮培養(yǎng)基,37℃5%CO2 培養(yǎng)。
三.一毫升小管操作方法 小管內(nèi)也是此細(xì)胞。
1. 用75%的酒精消毒(建議配制75%酒精的水是滅菌過(guò)的)。
2. 嚴(yán)格無(wú)菌操作,用吸管吸出管中細(xì)胞至9ml完全培養(yǎng)基混勻。
3. 1000rpm5min離心,重懸細(xì)胞。
4.換新的細(xì)胞培養(yǎng)瓶,37℃5%CO2 培養(yǎng)

帶急沖限制的加速功能為了降低機(jī)器機(jī)械零件磨損達(dá)到最佳加速,您可以在零件程序中選擇 SOFT 以確保連續(xù)、無(wú)振動(dòng)的加速。當(dāng)選擇“平穩(wěn)加速”時(shí),將以正弦曲線(xiàn)的形式生成路徑上的速度特性。
